I think that it is a sign of his ever-growing interest in her. He's trying to "feel her out" by asking her an almost teasing question. In fact, I think it's quite a daring one. The reel is a very fast, almost wild dance, not the type of thing you'd expect the conventional Darcy to participate in. I'm not sure what response he expected, but I have a feeling Elizabeth's pert answer pretty much fulfills his expectations. Hence his gallant rejoinder!
