It does seem that they know his exact age and are not just assuming that he's 35.

In the previous chapter, we read: "They would see, [Sir John] said, only one gentleman there besides himself; a particular friend who was staying at the park, but who was neither very young nor very gay."

It may be that Sir John's exact words, when he did tell them this, included Brandon's actual age and not that he was just 'not very young'.

I also can't see Col. Brandon announcing to the party that he's 35. But, on the other hand, Mrs. Jennings would not mind blurting something like this out.
