I don't think he is joking at Jane's illness, and I do not think Jane would be the butt of his joke if she were seriously ill. In fact, I think this line betrays his concern (to the degree that he ever shows such) for his daughter. He is belittling his wife (not his daughter), because Mrs Bennet does not take the illness seriously.
