Yes, I think Darcy knew that Bingley looked up to him and respected his opinion. He knew that his "assurances" would carry a lot of weight for Bingley.
This is why I think Darcy was wrong to meddle. Even if Jane were really indifferent, I think that the way he "advised" Bingley was wrong. He knows that Bingley will believe everything he says. Were Bingley of a different character, perhaps it would not have been so bad.
