The chapter starts out with Lydia asking the officers where Wickham is and why he's not there:

"...though this was not exactly the case[that Wickham had been excluded from the invitation], the absolute fact of his absence was pronounced by his friend Mr. Denny, to whom Lydia eagerly applied, and who told them that Wickham had been obliged to go to town on business the day before, and was not yet returned;"

So, since she started out asking about Wickham, it makes sense that she'd be the one with 'a thousand questions' for Caroline. It is out of character for Mary or Kitty to do this, and I agree that Caroline would not have spoken about Jane like that, nor would Jane have asked enough questions to annoy someone.
